Identification of leaf rust resistance genes in selected Argentinean bread wheat cultivars by gene postulation and molecular markers Electron. J. Biotechnol.
Vanzetti,Leonardo Sebastián; Campos,Pablo; Demichelis,Melina; Lombardo,Lucio Andres; Aurelia,Paola Romina; Vaschetto,Luis María; Bainotti,Carlos Tomás; Helguera,Marcelo.
Leaf rust, caused by Puccinia triticina Eriks. is a common and widespread disease of bread wheat (Triticum aestivum L.), in Argentina. Host resistance is the most economical, effective and ecologically sustainable method of controlling the disease. Gene postulation helps to determine leaf rust resistance genes (Lr genes) that may be present in a large group of wheat germplasm. Additionally presence of Lr genes can be determined using associated molecular markers. The objective of this study was to identify Lr genes that condition leaf rust resistance in 66 wheat cultivars from Argentina. Twenty four differential lines with individual known leaf rust resistance genes were tested with 17 different pathotypes of leaf rust collected from Argentina. Leaf rust...

Ecological setup, ploidy diversity, and reproductive biology of Paspalum modestum, a promising wetland forage grass from South America Genet. Mol. Biol.
Karunarathne,Piyal; Feduzka,Cristian; Diego,Hojsgaard.
Abstract With ever-rising demand for food, forage breeding for intensification of cattle production is also taking a leap. In South America, cattle production systems are displaced to marginal areas poorly exploited with cultivated pastures yet with high potential for growing stocking rates. This places the need for using native genetic resources to breed locally adapted plant genotypes that benefits from better forage quality, yield, and lesser threat to the local biodiversity. Paspalum modestum Mez is a grass species that produces quality forage and grows in marginal areas like estuaries and floodplains, suitable for introduction in breeding programs. In this study we characterize the species' reproductive biology and ecological preferences needed...

Expression profiles of sugarcane under drought conditions: Variation in gene regulation Genet. Mol. Biol.
Andrade,Júlio César Farias de; Terto,Jackeline; Silva,José Vieira; Almeida,Cícero.
Abstract Drought is a major factor in decreased sugarcane productivity because of the resulting morphophysiological effects that it causes. Gene expression studies that have examined the influence of water stress in sugarcane have yielded divergent results, indicating the absence of a fixed pattern of changes in gene expression. In this work, we investigated the expression profiles of 12 genes in the leaves of a drought-tolerant genotype (RB72910) of sugarcane and compared the results with those of other studies. The genotype was subjected to 80–100% water availability (control condition) and 0–20% water availability (simulated drought). To analyze the physiological status, the SPAD index, Fv/Fm ratio, net photosynthesis (A), stomatal conductance (gs) and...

Genome composition and pollen viability of Jatropha (Euphorbiaceae) interspecific hybrids by Genomic In Situ Hybridization (GISH) Genet. Mol. Biol.
Souza,Rosilda Cintra de; Marques,Daniela de Argollo; de Carvalho Filho,Marcel Mamede; Oliveira,Ana Rafaela da Silva; Siqueira,Walter José; Benko-Iseppon,Ana Maria; Brasileiro-Vidal,Ana Christina.
Abstract Interspecific hybridization is required for the development of Jatropha curcas L. improved cultivars, due to its narrow genetic basis. The present study aimed to analyze the parental genomic composition of F1 and BC1F1 generations derived from interspecific crosses (J. curcas/J. integerrima and J. curcas/J. multifida) by GISH (Genomic In Situ Hybridization), and the meiotic index and pollen viability of F1 hybrids. In F1 cells from both hybrids, 11 chromosomes of each parental was observed, as expected, but chromosome rearrangement events could be detected using rDNA chromosome markers, suggesting unbalanced cells. In the BC1F1, both hybrids had 22 chromosomes, suggesting that only n = 11 gametes were viable in the next generation. However, GISH...

Reciprocal recurrent selection effects on the genetic structure of tropical maize populations assessed at microsatellite loci Genet. Mol. Biol.
Pinto,Luciana Rossini; Vieira,Maria Lucia Carneiro; Souza Jr.,Claudio Lopes de; Souza,Anete Pereira de.
A modified reciprocal recurrent selection (RRS) method, which employed one cycle of high-intensity selection, was applied to two tropical maize (Zea mays L.) populations, BR-105 and BR-106, originating the improved synthetics IG-3 and IG-4, respectively. In the present study the effects of this kind of selection on the genetic structure of these populations and their synthetics were investigated at 30 microsatellite (SSR) loci. A total of 125 alleles were revealed. A reduction in the number of alleles was observed after selection, as well as changes in allele frequencies. In nearly 13% (BR-105) and 7% (BR-106) of the loci evaluated, the changes in allele frequencies were not explained, exclusively due to the effects of genetic drift. The effective...

Inclusion of genetic relationship information in the pedigree selection method using mixed models Genet. Mol. Biol.
Nunes,José Airton Rodrigues; Ramalho,Magno Antonio Patto; Ferreira,Daniel Furtado.
We used a mixed model approach and computer simulation to evaluate the inclusion of parentage information as determined by the genealogy established in the pedigree method. The simulations were based on a purely additive genetic model for one quantitative trait of 20 unlinked segregating loci with equal effects and an allelic frequency of 0.5 for heritability values of 10%, 25%, 50% and 75% for selection based on an F4:5 progeny mean. We simulated 1000 experiments for each heritability value, corresponding to the evaluation of 256 F4:5 progenies. The phenotypic values of the progenies were analyzed according to two models, one ignoring and one considering the additive genetic parentage among the progenies. The additive relationship coefficients among F4:5...

Characterization of biochemical compounds and antioxidant activity of “dedo-de-moça” chili pepper accessions Horticultura Brasileira
Gomes,Gisely Paula; Constantino,Leonel Vinicius; Corte,Ligia Erpen-Dalla; Riger,Cristiano Jorge; Chaves,Douglas S de A; Gonçalves,Leandro SA.
ABSTRACT Capsicum baccatum is one of the main chili pepper species grown and consumed in South America. In Brazil, C. baccatum var. pendulum is widely cultivated, especially “dedo-de-moça” type. For most horticultural crops, including Capsicum species their diversity remains under-explored for traits related to fruit quality, since over the past half century breeding efforts have focused mainly on good agronomic performances. The investment in screening traits such as flavor, nutritional compounds and antioxidant content in traditional varieties is very important to support fruit quality breeding efforts. Thus, the objective of this study was accessing the variations into content of soluble solid, acidity, capsaicinoid, vitamin C, total phenolic compounds...

Obtaining pepper F2:3 lines with resistance to the bacterial spot using the pedigree method Horticultura Brasileira
Riva-Souza,Elaine M; Rodrigues,Rosana; Sudré,Cláudia P; Pereira,Messias G; Viana,Alexandre P; Amaral Júnior,Antônio T do.
The economic importance of sweet pepper is rising in Brazil and abroad. Nevertheless, diseases, such as the bacterial spot (Xanthomonas campestris pv. vesicatoria or Xanthomonas euvesicatoria) jeopardize its yield. An efficient method for controlling the disease is the use of resistant varieties. The objective of this work was to select sweet pepper genotypes with resistance to the bacterial spot on a segregating population (Capsicum annum accessions UENF 1421, susceptible, x UENF 1381, resistant), advanced using the pedigree method. Three selection procedures, " selection between and within families" , " mass selection" , and " combined selection" were compared. Fifty-six F2:3 families (40 plants per family) were planted in the field, without replication....

Genetic dissimilarity, selection index and correlation estimation in a melon germplasm Horticultura Brasileira
Gomes,Danilo A; Alves,Igor M; Maciel,Gabriel M; Siquieroli,Ana Carolina S; Peixoto,Joicy Vitória M; Pires,Patrícia dos S; Medeiros,Iago A de.
ABSTRACT The success of breeding programs depends on genetic variability. Individuals selected based on a few traits may be a limitation. One alternative is the use of nonparametric indices. However, there is no information on the use of selection indices in melon germplasms. The present study aimed to estimate genetic dissimilarity in a melon germplasm and select potential parent plants for future breeding programs. The genetic material consisted of 37 melon accessions. The traits assessed were fruit diameter and length, diameter and length of the fruit cavity and total soluble solids. Genetic dissimilarity was assessed by multivariate analyses (UPGMA and Tocher). Selection gain estimates were analyzed by comparing the classic Smith-Hazel and sum of ranks...

Resistance sources to root-knot nematodes Meloidogyne javanica, M. incognita and M. enterolobii in sweet potato Horticultura Brasileira
Carmona,Paula Andrea O; Pinheiro,Jadir B; Amaro,Geovani Bernardo; Silva,Giovani Olegario da; Peixoto,José Ricardo; Cares,Juvenil Enrique.
ABSTRACT One of the main obstacles for food production in many developing countries, as in Brazil, is the damage caused by root-knot nematodes, mainly those belonging to the genus Meloidogyne. This study aimed to assess the resistance levels of 44 sweet potato genotypes to M. javanica, M. incognita race 1 and M. enterolobii. These researches were carried out in 2014, under greenhouse conditions in Brasília-DF, Brazil. A completely randomized design with six replicates of one plant/plot/treatment was used. We determined the gall index (GI) and egg mass index (EMI) in the root system of each plant, the number of eggs and juveniles per gram of root with galls and the nematode reproduction factor. M. javanica was less aggressive and reproduced in only 9.09% of...

Genetic improvement of horticultural crops mediated by CRISPR/Cas: a new horizon of possibilities Horticultura Brasileira
Koltun,Alessandra; Erpen-Dalla Corte,Lígia; Mertz-Henning,Liliane Marcia; Gonçalves,Leandro SA.
ABSTRACT The burden of the current global challenge involving food security lies in the need to improve crop production. In this regard, biotechnology stands out as an essential tool to generate plants able to cope with pests, diseases, and harsh climatic conditions, and more efficient in the use of natural resources. An advanced approach to create genetic variability in a precise and targeted way, the genome-editing technique CRISPR/Cas (clustered regularly interspaced short palindromic repeats/CRISPR associated proteins), has drawn the attention of breeders. The genome editing CRISPR/Cas system relies on a guiding RNA that directs a nuclease to generate a double-strand break (DSB) at a target DNA, activating the cell repair systems and eventually leading...

Resistance of tomato genotypes to the silverleaf whitefly mediated by acylsugars Horticultura Brasileira
Resende,Juliano TV de; Maluf,Wilson R; Cardoso,Maria das Graças; Gonçalves,Luciano Donizete; Faria,Marcos V; Nascimento,Ildon R do.
Acylsugars in the leaflets of the wild accession LA-716 (Lycopersicon pennellii) gives resistance to Bemisia spp., which causes expressive losses to the tomato crop. The goal of this work was to evaluate the resistance to the silverleaf whitefly (Bemisia tabaci biotype B) of previously selected genotypes, based on the level of acylsugars in the leaflets. F2BC1 plants {F2[Lycopersicon esculentum TOM 584 x (L. esculentum TOM 584 x L. pennellii LA-716)]} were selected for extreme contents of acylsugars: BPX-370B pl#30-275, BPX-370B pl#79-278, BPX-370B pl#30-380, BPX-370B pl#25-271 (high contents), BPX-370B pl#30-02, BPX-370B pl#30-142 (low contents). These genotypes, along with the parents TOM 584 and LA-716, were submitted to a silverleaf whitefly...
